     Effects of Seed Priming on Germination and Seedling Growth of Radix Isatidis under Drought Stress

     In order to screen out the appropriate seed initiator for Radix Isatidis,four initiators including PEG-6000,CaCl2,GA3 and H2O were used to treat the Radix Isatidis seeds,and then the seed germination and seedling growth characteristics of Radix Isatidis were investigated under 20% PEG-6000 simulated drought stress.The results showed that all these four initiators could improve seed germination rate, germination potential and germination index in different degrees under drought stress.The 18 h water priming had the best effect on seed germination,and the seed germination rate,germination potential,and germination index were increased by 33.3%,36.6% and 106.4% respectively compared with CK2(drought stress and no initiators).The follower was 25% PEG-6000 priming,and the germination rate,germination potential,germination index were 35.2%,51.2%,and 32.8% higher than CK2,respectively.The 50 mg/L GA3 priming had a significantly higher effect than other treatments on germination index,which was increased by 171.1% compared with CK2.The 1.5% CaCl2 had lowest effect on germination rate, germination potential and germination index,which only were increased by 16.7%,19.5% and 23.9% respectively compared with CK2.In addition,after seed priming,the plant height,root length,chlorophyll content of seedlings were significantly increased,and the relative conductivity was decreased.In conclusion,the seed priming effect was in turns: 18 h water >25% PEG-6000 >50 mg/L GA3 >1.5% CaCl2 ,which suggested that 18 h water priming to treat the seeds before sowing could greatly improve the seed germination and seedling growth of Radix Isatidis

    Effects of Different Reagents and Temperature on Removing Seed Dormancy of Paris polyphylla var.yunnanensis

    The effects of different concentrations of gibberellin(GA),fluridone(FL) to soak the seeds and different stratification temperature on seed germination and embryo rate of Paris polyphylla var.yunnanensis were studied respectivly to screen the best way to revive the seed of Paris polyphylla var.yunnanensis.The results showed that GA treatment alone,GA+FL combination and sand storage at constant temperature of 20℃ could effectively break the dormancy of the seeds.Among them,the best germination was at 300 mg/L with GA treatment alone, and the germination rate and germination potential were 79.00% and 54.33% respectively.The highest germination rate was 81.33% with GA+FL combination(300 mg/L GA+10 mg/L FL),and the highest germination potential was 54.33% with 300 mg/L GA+0 mg/L FL,and the rotten seed rate was 4.33% with 200 mg/L GA+10 mg/L FL.The seed embryo rate of Paris polyphylla var.yunnanensis increased fastest under constant temperature of 20 ℃ in different temperatures with sand storage.By all accounts of the seeds germination of Paris polyphylla var.yunnanensis,the effects of GA+FL combination and constant temperature of 20℃ sand storage were better than other treatments.That is to say,300 mg/L GA+10 mg/L FL is the optimal seed soaking combination,and constant temperature of 20℃ is optimal for sand storage.

    Effects of Soaking Seeds with Salicylic Acid on Seed Germination and Drought Resistance of Dracocephalum tanguticum

    In order to investigate the effects of exogenous salicylic acid on germination and drought stress of Dracocephalum tanguticum,different concentrations of SA(0,0.2,0.4,0.6,0.8 mmol/L) were used to soak the seeds of D.tanguticum,and then the seed germination and seedling physiological characteristics of D.tanguticum were studied under 20% PEG-6000 simulated drought stress.The results showed that 0.6 mmol/L SA had the best effect on germination by soaking seed,the germination rate,germination energy,germination exponent and vigor index were increased by 34.82%,34.42%,34.87% and 54.86% respectively compared with CK;Under drought stress,the exogenous SA increased the content of proline and decreased the malondialdehyde content at the concentration of 0.6 mmol/L,however,the chlorophyll and soluble protein contents were similar compared with CK.In conclusion,exogenous SA could improve the seed germination by promoting germination energy,germination exponent and vigor index,and increase the ability of D.tanguticum to resist drought stress by enhancing the ability of plants to resist osmotic stress,reducing the cell membrane damage,and 0. 6 mmol/L SA soaking for 24 h was the best.

    Effects of Plant Growth‑Promoting Bacteria Containing ACC Deaminase on Seed Germination of Psammosilene tunicoides

    The effects of plant growth‑promoting bacteria containing ACC deaminase on seed germination of Psammosilene tunicoides were studied,at the same time,the most suitable growth‑promoting strain was screened out to provide theoretical basis for the development and utilization of microbial fertilizer and high efficient cultivation of P.tunicoides.Four PGPB strains containing ACC deaminase and their mixed strains were used to impregnate P.tunicoides seeds and accelerate their germination.The germination rate,germination energy,germination index and daily average germination rate and so on were measured with sterile water as control.The results showed that,different PGPB strains containing ACC deaminase showed different effects on seed germination of P.tunicoides,among which F195 strain had the best germination effect. Compared with CK,seed germination rate,germination energy,germination index,daily average germination rate and vigor index increased by 32%,49%,223%,32% and 358%,respectively.The germination days of the first seed and peak days were advanced by 3 days and 2 days respectively,the minimum of average germination days was 8.30 d.The percentage of water absorption of seeds increased by 80%. Root length,germ length,seedling fresh weight and seedling dry weight increased by 63%,63%,76% and 131%,respectively.Z24 and Mix strains treatments had better germination effect,which were significantly different from CK(P<0.05).The results of principal component analysis showed that the germ length,root length,germination rate,daily average germination rate and vigor index were higher in component 1,which could be used as important indexes to evaluate the germination and growth of P.tunicoides seeds under different PGPB strains.Cluster analysis showed that F195 strain had best germination effect,followed by Z24,Mix and Z39 strains.In conclusion,F195 strain has good growth‑promoting characteristics during seed germination.
