黄淮肉羊CIDEa 和CIDEc 基因组织表达与肌内脂肪含量的相关性分析

Correlation Analysis between Tissue Expression of CIDEa and CIDEc Genes and Intramuscular Fat Content in Huanghuai Sheep

摘要: 为了探究CIDEa CIDEc 基因在黄淮肉羊各组织的表达规律,并分析其与肌内脂肪及脂肪酸含量的相关性,选取3月龄、9月龄、18月龄的黄淮肉羊各3只,屠宰后采集各组织样品并测定其肌内脂肪和脂肪酸含量,利用RT-qPCR技术检测其在不同组织以及不同月龄背最长肌中的表达水平,利用皮尔逊相关系数分析其与肌内脂肪和脂肪酸含量的相关性。结果表明,CIDEaCIDEc在皮下脂肪和腹脂中均具有较高的表达水平。CIDEa在4种肌肉组织(半腱肌、背最长肌、臂三头肌、股四头肌)的表达量总体上均随着年龄的增加而增多;CIDEa在背最长肌中的表达量与肌内脂肪、棕榈酸(C16:0)含量均呈正相关(P<0.05),与硬脂酸(C18:0)、花生二烯酸(C20:2)、花生四烯酸(C20:4n6)含量均呈负相关(P<0.05)。而CIDEc 基因在背最长肌和臂三头肌中的表达量总体上随着年龄的增加而降低。CIDEc与十三碳酸(C13:0)、硬脂酸(C18:0)、花生二烯酸(C20:2)、二十三碳酸(C23:0)、二十四碳酸(C24:0)含量均呈正相关(P<0.05);与银杏酸(C17:1)、油酸(C18:1n9c)含量均呈负相关(P<0.05)。综上,CIDEaCIDEc基因在黄淮肉羊肌内脂肪和脂肪酸调控方面具有重要作用。

Abstract: The aim of this study was to explore the expression patterns of CIDEa and CIDEc genes in various tissues of Huanghuai sheep,and analyze their correlation with intramuscular fat and fatty acid content. This study selected three male Huanghuai sheep at the ages of 3,9,and 18 months each,collected tissue samples after slaughter and measured their intramuscular fat and fatty acid content.It used RT⁃qPCR technology to detect CIDEa and CIDEc genes expression level in various tissues and in longissimus dorsi at different ages,and analyzed their correlation with intramuscular fat and fatty acid content using Pearson correlation coefficient.The results showed that both CIDEa and CIDEc had highest expression levels in subcutaneous fat and abdominal fat.The expression level of CIDEa in 4 muscle tissues semitendinosus,longissimus dorsi,triceps brachii and quadriceps femoris increased overall with age.The expression level of CIDEa in the longissimus dorsi was positively correlated with intramuscular fat and C16:0(P<0.05),and negatively correlated with C18:0,C20:2,and C20:4n6(P<0.05). While the expression level of CIDEc gene in longissimus dorsi and triceps brachii decreased with age.The expression level of CIDEc in the longissimus dorsi was positively correlated with C13:0,C18:0,C20:2,C23:0,and C24:0(P<0.05),and negatively correlation with C17:1 and C18:1n9c(P<0.05).In summary,CIDEa and CIDEc genes play important roles in the regulation of intramuscular fat and fatty acids in Huanghuai sheep.
