玉米HD-Zip转录因子基因ZmHOX32的克隆及功能探究

Cloning and Function Analysis of Maize HD-Zip Transcription Factor Gene ZmHOX32

摘要: 同源异型域-亮氨酸拉链蛋白(HD-Zip)可通过与其他蛋白质互作参与激素信号传导途径,进而调控植物对逆境胁迫的应答和生长发育等过程。为验证及探究HD-Zip蛋白对干旱胁迫和ABA诱导的响应模式,从玉米抗旱自选系郑D58M中克隆了1个HD-Zip转录因子基因,暂时命名为ZmHOX32。ZmHOX32蛋白包含856个氨基酸,属于亲水性蛋白质,定位于细胞核内。进化树和保守基序分析发现,ZmHOX32蛋白序列与谷子、胡桃木的同源蛋白序列高度相似且保守基序完全一致。ZmHOX32基因启动子序列含有响应激素、非生物胁迫及光刺激的结合元件。qRT-PCR结果显示,ZmHOX32基因属于组成型表达基因,在营养分生组织中高表达,且受干旱胁迫和外源ABA的正向诱导,表明ZmHOX32基因参与干旱胁迫和ABA信号传导途径。

Abstract: The homeodomain-leucine zipper protein(HD-Zip) can participate in the hormone signaling pathway by interacting with other proteins,thereby regulating the stress response and growth and develop ment of plants.To verify and explore the response pattern of HD-Zip protein to drought stress and ABA in duction,a HD-Zip transcription factor gene was cloned from maize drought-resistant self-selection line Zheng D58M,which was temporarily named ZmHOX32.The ZmHOX32 protein consisted 856 amino acids and belonged to a hydrophilic protein,which was localized in the nucleus.Phylogenetic tree and conserved motif analysis showed that the ZmHOX32 protein sequence was highly homologous to Setaria italica and Panicum hallii protein sequences,and the conserved motif was completely identical.The ZmHOX32 gene promoter sequence contained binding elements that responded to hormones,abiotic stresses and light stim ulation.The results of qRT-PCR showed that ZmHOX32 gene was a constitutively expressed gene,which was highly expressed in vegetative meristem and was positively induced by drought stress and exogenous ABA.These results indicate that ZmHOX32 gene is involved in drought stress and ABA signaling path way.
